Abstract :

ABSTRACT This study aims to determine the time of immunization antibody titer indicated by the value of optical density (OD). This research was using adult male rabbits of local strain. Spermatozoa in the cauda epididymis was collected by flushing. Furthermore, isolated sperm membrane proteins were in solution hypoosmotic (10 mM potassium buffer phosphate) with centrifugation techniques. Rabbits immunized with the sperm membrane protein dose 200 mcg/pi. Immunization was done 3 times with an interval of 21 days. Antibody titer tested by ELISA. Negative control using rabbit immunized with no antigen. OD value of the data were analyzed with anova and LSD test. The results showed that the average antibody titer after immunization II (1.777) and III (1.853) higher than after immunization I (1.490) and prior immunization / pre-immune (0.652). The conclusion of this research is a long time effect on immunization antibody titer.
