Extracellular signal regulated kinase (erk1/2), reseptor estrogen (er) a, and insuline like growth factor i (igf-ir) expressions on osteoblast in influencing bone density in the female young rat (rattus norvegicus) after exercise training

Abstract :
It is known that exercise training can increase bone density both for male and female on human and animals. The fact shows that increase bone density mechanism by exercise training up to know is not yet clear. Exercise training in the young is very important because one of the main cause factors of osteoporosis is peak reach of bone density that is not optimal in the young. It can cause bone strength decreasing. This research was aimed to know the effect of exercise training on moderate intensity on ERα, ERK1/2, and IGF-IR expression and it’s role in influencing bone density. This research was The Randomized Posttest only Control Group Design. The experimental unit in this research was white rat strain wistar, female in the age of 4 moths, and total number were fourteen. Independent variables of this research was exercise training on moderate intensity, and dependent variables were ERα, ERK1/2, and IGF-IR expression and bone density. ERα, ERK1/2, and IGF-IR expression was measured by using immunobloting methode, while bone density was measured by using ultra-sound DBM 1200. The data analysis used was MANOVA and Pearson correlations. Base on MANOVA analysis showed that there were increasing of ERα, ERK1/2, and IGF-IR expression and bone density together on exercise training (p = 0.00 < α = 0.05). Based on Pearson correlation analysis showed that increasing ERα, ERK1/2, and IGF-IR give contribution in increasing bone density r = 0.692 for ERα, r = 0.711 for ERK1/2, and r = 0.608 for IGF-IR (αααααααααααααααααααααααα= 0.05). Futhermore a strong correlation was found between ERα and IGF-IR r = 0.965 and EKR1/2 r = 0.985 also between IGF-IR and ERK1/2 r = 0.914 (ααααααααααααααααα= 0,05). ..............There fore this study concluded that exercise training be able to increase bone density through ERK1/2-ERα-IGF-IR pathway.
