Identification of fertilization receptor protein (zp3) on goat zona pellucida as candidate of immunocontraceptive substance using in vitro fertilization technique

Abstract :
he researches of immunocontraception have been done in several species, but haven’t yet been done in goat. As well as known thathormonal contraception up to now is still having side effects. In preliminary study, a crude of goat zona pellucida protein was effectiveprohibitted graviditation of mice. The aim of this study was to prove that obtained gZP3 protein isolate was representing receptor ofgoat sperm. Isolate of gZP3 was incubated on goat sperm, while antibody of gZP3 was incubated on goat oocyte, then each of them wasin-vitro fertilized separately. Antibody of gZP3 produce on rabbit (Oryctolagus cuniculus). The first immunization, gZP3 suspensionwas mixed with Complete Freund’s Adjuvant (CFA) 1:1 (v/v) followed by boosters twice with the interval of 11 days, the suspension wasmixed with Incomplete Freund’s Adjuvant (IFA) 1:1 (v/v). In in-vitro fertilization technique, antibody of gZP3 which was supplementedmedia during maturation process of goat oocytes in vitro had decreased (p < 0.05) embryos cleavage rate. Protein of gZP3 which wassupplemented in the capacitation media of goat sperms decreased (p < 0.05) of cleavage rate. Based on these results, it was concludedthat antibody gZP3 having a fertilization receptor protein at the goat zona pellucida.
