Influent of e. coli and leucocytes againts human sperm quality in vitro

Abstract :
Male genital tract infection has been recognized as one of the causes of infertility. Indicators of the occurrence of genital tract infection is the presence of bacteria during semen culture (bacteriospermia) and the finding of leucocyte of more than 1 million/ml semen (leucotytospermia). E. coli is the most common cause of prostatitis and epididymitis. The objective of this study was to explain the effect of E. coli and leucocyte on spermatozoa quality in vitro. This study comprised three experiments. In experiment 1 sperm was incubated with E. coli, in experiment 2 with lymphocyte, and experiment 3 with granulocyte. In those experiments, sperm motility and vitality. Spermatozoa were obtained from donor with normal spermatozoa according to WHO (1999). E. coli was obtained by culturing the semen of infertile males. Lymphocyte and granulocyte were obtained from donors’ blood. Sperm preparation was made using Percoll gradient column method. Lymphocyte isolation was performed using Histopaque 1077, and granulocyte isolation used Histopaque 1077 and 1119. Sperm motility and vitality was observed by means of conventional method with microscope. The result of this study showed that in vitro E. coli had effect on the motility (p = 0.000) and vitality(p = 0.000) of sperm. Lymphocyte had effect on the motility (p = 0.034) and vitality (p = 0.041). Granulocyte had effect on motility (p = 0.000) and vitality (p = 0.017). In conclusion, E. coli has effect on the reduction of sperm quality through E. coli adhesion to the sperm. In leukocyte, granulocyte has higher effect in the reduction of sperm quality. Sperm quality reduction by granulocyte undergoes through the mechanism of ROS production.
