Abstract :
Corn cobs have much hemicellulose and cellulose which can be hydrolyzed and fermented become bioethanol, but corn cobs have a lignin which can inhibits hydrolysis process, so must require pretreatment to degradation of lignin. The purpose of this research is got the maximum lignin degradation by mixing between the enzymatic and chemical processes. This research will be used NH4OH, H2O2, laccase with NH4OH, and laccase with H2O2 for the pretreatment of substrate. Based on analysis, the lowest residual lignin of corn cobs in pretreatment with NH4OH 15%. In this research, laccase was able to degrade partially lignin in substrate but also able to degrade partially hemicellulose, so can be reduced of monosacharide in substrate. Substrate from pretreatment NH4OH 15% hydrolyzed by enzyme cellulase, xylanase, and xylose isomerase with ratio activity of enzyme 1:1:1 for 6 hours and produce glucose and xylulose. Hydrolyzate of the process enzyme hydrolysis is fermented by the yeast Saccharomyces cereviceae for 3 days. Levels content of fermentation’s product called ethanol are calculated using the method of Skoog which based on the density of ethanol. These results indicate that levels content of ethanol fermentation about 5.66% (v/v).
