The effect of antibodies anti-inhibin in obtaining of anti-idiotipic antibodies on mice

Abstract :

Forty Balb/c mice 8 weeks old were divided into 4 groups. Mice in group-I were injected with antibody/anti-inhibin 1:10 0.2 ml sc; Mice in group-II were injected twice by using the same dosage antibody/anti-inhibin; Mice in group-III were injected three times; and Mice in group-IV were injected four times. Five male rabbits aged between 6 and 7 month were used in this study to produce antibody/ anti-inhibin. This injection was done once in 2 weeks and before treatment all of the mice were oestrous synchronized with 25 mg PMSG im. The level of antibody/anti-idiotypic and the examination of antigenic similarity between inhibin and idiotypic antibody were measured by using indirect ELISA. While the level of total immunoglobulins were measured by using direct ELISA. The data were analyzed by Anova for antibody anti-idiotypic and total immunoglobulin, and t test for antigenic similarity. The result showed that injection of rabbit antibody/anti-inhibin repeatedly stimulated the use of mice antibody/anti-idiotypic. Antigenic similarity have similar between epitop inhibin with idiotype of mice antibody/anti-idiotypic, based on reaction of rabbit antibody/anti-inhibin, beside that finding it is also known that antibody anti-inhibin may decreased the total immunoglobulins of mice.
