Abstract :
The research purpose was to determine the optimation of tyrosine kinase activity from sperm isolation of FH cow according to pH, temperature and optimum incubation time. Fresh semen as samples were collected using artificial vagina. Tyrosin kinase was isolated by means of centrifugation and ultrasonication. Activity unit of cow sperm tyrosine kinase is the amount of fosfat µmol of transferred ATP by each mililiter tyrosine kinase enzyme per minute in optimum condition. The optimation of tyrosine kinase activity of sperm according to optimum condition included different pH (6, 6.5, 7, 7.5, 8), temperature ( 20, 25, 30, 35, 40)oC and incubation time (20, 25, 30, 35, 40) minute activity was optimum. The result of this research showed that the optimum activity tyrosine kinase of sperm at pH 7, temperature 35oC and 30 minutes incubation time with 0,0219 unit activity.
