The effect of fat tissue mesenchymal stem cells to the total amount of receptor of basic fibroblast growth factor (bfgf) in wound healing process on males rabbit (oryctolagus cuniculus)

Abstract :

Backgrounds : Nonhealing wounds remain a significant challenge for plastic surgeon. Treatment of chronic wounds remains difficult, in spite of better understanding of pathophysiologic and greater adherence to recognized standards of care. Even with recent advances stemming from breakthroughs in recombinant growth factors, up to almost 50% of chronic wounds that have been present for more than a year remain resistant to treatment. Adipose- derived stem cells (ADSCs) are adult stem cells that are easily harvested and of great interest for plastic surgeon. Mesenchymal stem cells within the stromal- vascular fraction of subcutaneous adipose tissue, that adipose- derived mesenchymal stem cells have been used in skin repair with satisfactory results. The production and secretion of growth factors have been reported to be an essential function of ADSCs. Specifically, ADSCs secrete basic Fibroblast Growth Factor (bFGF)’s reseptors that can induce tissue regeneration and stimulates healing of skin wounds rapidly and perfectly formed as function and aesthetics.   Objective : to analyze the role of mesenchymal stem cells of fat tissue to the amount of basic fibroblast growth factor (bFGF/FGFR2) receptors,  which probably has the role of accelerate the wound healing process Design : experimental research Methods : Experimental study using post test only control group design. 18 male rabbits oryctologus cuniculus at about 3 months old will be used. Three 1,5cmx1,5cm full thickness excision will be made at each rabbit’s right and left back. The right wounds(TG, n=18)  treated with adipose- derived mesenchymal stem cell topically cells 2x 106 cell count on day-0. The left wounds (CG, n=18)  treated moist. Skin specimens will be collected on the 3rd,14th,21st days from 6 different rabbits at each period. The sections will be stained with antibody FGF-2  for examining the number of basic Fibroblast Growth Factor’s reseptor with immunohistochemical staining. The data will then be analyzed statistically using ANOVA (Analysis of Variant).   Result : On all of the wounds in which were applied topical adipose- derived mesenchymal stem cells, the number of basic Fibroblast Growth Factor’s reseptor at inflammatory phase, proliferation phase and maturation phase significantly higher with respect to the control (p< 0,0001) and (p = 0,010).   Conclusion : mesenkimal stem cells topically have an influence on the amount of FGF receptors basis through transdifferentiation mechanism parakrin equal to signal that causes wound healing is faster and perfect both functional and aesthetic.
