Abstract :
The aim of this study was to establish an optimal marker set for a routine paternity and identity test. In the present work a paternity and identity test within three pig breeds (DE, DL and Pi) was performed by means of microsatellite analysis. Genotyping of the animals was done after PCR amplification of the alleles on an automated DNA sequencer (LI-COR, DNA Analyzer, GENE READIR 4200 (MWG BIOTECH). For the examination of the applicability of the microsatellite markers in paternity test, the exclusion probabilities and Polymorphism Information Contents (PICs) were calculated. Combined exclusion probability over 99 % has been achieved in all breeds by using 8 microsatellites. Fifteen of the 25 loci were selected and a group of 5 loci were packed together in three multiplex-PCRs as possible marker sets for a paternity and identity test in pig.
